The squadron was first activated in 1943, during World War II as the 756th Bombardment Squadron. After training in the United States, it deployed to the Mediterranean Theater of Operations, Torre Giulia Field, Italy where it participated in the strategic bombing campaign against Germany, and it earned a Distinguished Unit Citation for its actions. Following V-E Day, the squadron returned to the United States and was inactivated.
In 1947, the squadron was reactivated in the reserves, but does not appear to have been fully manned or equipped before inactivating in June 1949. It was again activated in the reserves a few months later as a corollary unit, flying with the active duty 301st Bombardment Group. It was called to active duty for the Korean War, but did not serve as a unit, its personnel being used as fillers for other units, while the squadron was once again inactivated.
The squadron was re-designated the 756th Troop Carrier Squadron and activated at Andrews in the reserve in 1954 flying the C-46 and C-119. The unit later converted to the 756th Military Airlift Squadron flying the C-124. After, 756th Tactical Airlift Squadron flying the C-130 and 756th Military Airlift/756th Airlift Squadron flying the C-141, respectfully until 2003. The unit once again converted to its current role, flying KC-135's supporting the air refueling mission, as the 756th Air Refueling Squadron.

The Association was originally formed within the State of Maryland on October 12, 1989 as the 756th Squadron Association. The association thrived until a motion was passed to open the association to any member in the 459th wing. As more 459th members joined, the motion was passed to consider changing the name of the association. On December 14, 2010, the Board of Directors submitted a name change of the Association to the 459th Wing Association, as such, dissolving the 756th Squadron Association, and along with it, its original purpose to promote esprit de corps among former members of the 756th squadron heraldry.
To correct this misstep, former members of the 756th came together to reinstate the association.
On June 11, 2020, the 756th Squadron Association was re-established and formed in the State of Virginia.

The 756th ARS is flying the KC-135R, "Stratotanker" with a crew of three aircrew members: Pilot, Co-pilot & Boom Operator
The 756th ARS provides unsurpassed Air Refueling, supporting Aeromedical Operations platforms, and Agile Combat troop and cargo movements from our Nation's Capital to the World.
The KC-135R has been modified with the new CFM International (CFM56)
(Military designation: F108) high-bypass turbofan engines produced by General Electric and Safran. The CFM56 engine produces approximately 22,500 lbf (100 kN) of thrust, nearly a 100% increase compared to the original J57 engine. The modified tanker, designated KC-135R (modified KC-135A or E) or KC-135T (modified KC-135Q), can offload up to 50% more fuel (on a long-duration sortie), is 25% more fuel-efficient, and costs 25% less to operate than with the previous engines. It is also significantly quieter than the KC-135A, with noise levels at takeoff reduced from 126 to 99 decibels.
The KC-135R's operational range is 60% greater than the KC-135E for comparable fuel offloads, providing a wider range of basing options.
